A quantitative in vivo assay for craniofacial developmental toxicity of histone deacetylases

چکیده
Many bony features of the face develop from endochondral ossification preexisting collagen-rich cartilage structures. The proper development these structures is essential to morphological formation face. developmental programs governing pre-bone facial cartilages are sensitive chemical compounds that disturb histone acetylation patterns and chromatin structure. We have taken advantage this fact a quantitative assay craniofacial toxicity based on distortion deterioration in zebrafish larvae upon exposure increasing concentrations several well-described deacetylase inhibitors. In assay, we measure angle formed by developing ceratohyal bone as precise, proxy for overall status cartilages. Using well-established toxicant deacetylase-inhibiting compound valproic acid along with 12 structurally related compounds, demonstrate applicability investigate structure-activity relationships.
